To clarify the fixing, does the base fit to the door independently of the knob with the square spindle fitting through the base and the knobs fitting either side with a grub screw fixing the knob to the bar which is then turned to activate the mortice latch.

Yes, your understanding is correct. The base of the door knob fits independently to the door. The square spindle passes through the base, allowing the knobs to be mounted on either side. Each knob is secured to the spindle with a grub screw. When the knob is turned, it activates the mortice latch as intended. I hope that makes sense!

Is the rose on the brass option a screw on one or is it fixed some other way?

The rose is screwed onto the door and the knob just sits on top.
